When Lucky Luciano
and  fellow mobsters Meyer Lanski, Frank Costello, Joe Adonis, Lepke Buchalter, and Dutch Schultz,  set up a meeting to discuss the formation of a National Crime Syndicate to end the reign of the Mustache Petes, and reduce rival gang conflicts.  Vincent Mangano was one of the few old school gangsters to join forces with Luciano and the others, going as far as helping him eliminate some of the old bosses.  Luciano appointed Mangano as boss of would later be known as the Gambino Family; he trusted Mangano to run his affairs while he served time in jail for vice charges.  

Vincent Mangano, his brother Philip, and Albert Anastasia were members of Murder Inc., a notorious contract killing organization believed to be behind a large number of mob hits. Anastasia and Mangano would have frequent arguments during meetings, arguments that would often turn to violence.

Mangano had many enemies. Anastasia,  Frank Costello, Joe Adonis, and Thomas Lucchese had all expressed a desire to rub out the old school boss, and to this day it isn't clear what exactly happened to Vincent Mangano.  His brother Philip was found dead in a field, naked from the waist down, and the police, wanting some answers, went to visit Vincent, only to realise that he too had disappeared.  While no body was ever found, it is believed Mangano met with foul play.

Shortly thereafter, Albert Anastasia took over as boss...
